Detecting and treating breast cancer resistance to EGFR inhibitors

التفاصيل البيبلوغرافية
العنوان: Detecting and treating breast cancer resistance to EGFR inhibitors
Patent Number: 9,303,088
تاريخ النشر: April 05, 2016
Appl. No: 13/248690
Application Filed: September 29, 2011
مستخلص: The application describes therapeutic compositions and methods for treating cancer. For example, therapeutic compositions and methods related to inhibition of FAM83A (family with sequence similarity 83) are provided. The application also describes methods for diagnosing cancer resistance to EGFR inhibitors. For example, a method of diagnosing cancer resistance to EGFR inhibitors by detecting increased FAM83A levels is described.

Claim: 1. A method of predicting a clinical outcome for a patient having breast cancer, comprising: providing a biological sample comprising breast cancer cells from the patient; contacting said breast cancer cells with an antibody specific for FAM83A; detecting a FAM83A expression level in the breast cancer cells by measuring the level of binding between the antibody and the breast cancer cells; and predicting the clinical outcome of the patient based on the FAM83A expression level, wherein the FAM83A expression level is positively correlated with the breast-cancer related mortality.
Claim: 2. The method of claim 1 , wherein predicting the clinical outcome comprises generating a Kaplan-Merier survival curve.
Claim: 3. The method of claim 1 , wherein the antibody specific for FAM83A is a polyclonal antibody against FAM83A.
Claim: 4. The method of claim 3 , wherein the polyclonal antibody is raised against a peptide having an amino acid sequence of SEQ ID NO: 6.
Claim: 5. The method of claim 3 , wherein the polyclonal antibody is raised against a peptide having an amino acid sequence of SEQ ID NO: 2.
Claim: 6. The method of claim 1 , wherein the antibody specific for FAM83A protein is a monoclonal antibody against FAM83A.
Claim: 7. The method of claim 1 , wherein high expression level of FAM83A in said patient is indicative of high clinical mortality as compared to patients carrying tumors with lower expression levels of FAM83A.
Claim: 8. The method of claim 1 , wherein the biological sample is a biopsy sample from the patient.
