The present invention relates to processes for preparing carboxylated latex particles based on the copolymerization of a monoalkenylaromatic monomer (A) and an aliphatic unsaturated carboxylic acid (B), where the copolymerization takes place at a pH in the range from pH >1.5 to pH <4.5. The pH is typically monitored during the copolymerization process and regulated by suitable compensatory titration. The invention likewise relates to the carboxylated latex particles obtainable by one of the processes according to the invention, and to the use of these particles in an immunological test method. |

Claim: |
1. A process for preparing carboxylated latex particles comprising the steps of: forming a mixture comprising a monoalkenylaromatic monomer and an aliphatic unsaturated carboxylic acid, whereby copolymerization and carboxylation of the monomer takes place and wherein the mixture has a pH between 1.5 and 4.5, monitoring the pH during copolymerization, and regulating the pH by compensatory titration so that it remains constant during copolymerization, thereby forming carboxylated latex particles. |
Claim: |
2. The process of claim 1 wherein the monoalkenylaromatic monomer is selected from the group consisting of styrene and alpha-methylstyrene. |
Claim: |
3. The process of claim 1 wherein the aliphatic unsaturated carboxylic acid is selected from the group consisting of acrylic acid, methacrylic acid, fumaric acid, and itaconic acid. |
Claim: |
4. The process of claim 1 wherein the mixture is an aqueous emulsion. |
Claim: |
5. The process of claim 1 wherein the mixture further comprises a water-soluble peroxide. |
Claim: |
6. The process of claim 1 wherein the mixture further comprises a seed latex. |
Claim: |
7. The process of claim 1 wherein the aliphatic unsaturated carboxylic acid is dissolved in the monomer prior to forming the mixture. |
Claim: |
8. The process of claim 1 wherein the pH of the copolymerization mixture deviates from a target pH by a maximum of ±0.3. |
Claim: |
9. The process of claim 1 wherein the regulation step utilizes an aqueous solution of an alkali metal carbonate, bicarbonate, or hydroxide as a regulating reagent. |
Claim: |
10. The process of claim 1 wherein the pH is greater than or equal to 2.0 and less than 4.0. |
Claim: |
11. A carboxylated latex particle prepared by a process comprising the steps of: forming a mixture comprising a monoalkenylaromatic monomer and an aliphatic unsaturated carboxylic acid, whereby copolymerization and carboxylation of the monomer takes place and wherein the mixture has a pH between 1.5 and 4.5, monitoring the pH during copolymerization, and regulating the pH by compensatory titration so that it remains constant during copolymerization, thereby forming carboxylated latex particles. |
Claim: |
12. A conjugate comprising a particle according to claim 11 and a biomolecule selected from the group consisting of nucleic acids, peptides, proteins, haptens, lipids, and polysaccharides. |
Claim: |
13. An immunological test method to determine an antigen or antibody comprising the steps of combining a sample containing the antigen or antibody with a conjugate comprising a particle according to claim 11 and a binding partner for the antigen or antibody whereby the particle aggregates with the antigen or antibody and detecting aggregation of the particle as a measure of the antigen or antibody in the sample. |
