مستخلص: Guanylyl Cyclase-1 (GC-1), or soluble guanylyl cyclase (sGC), is a heterodimeric heme-containing protein that responds to the binding of nitric oxide (NO) with enhanced cyclase activity, producing cyclic guanosine monophosphate (cGMP) and a signaling cascade. NO regulates numerous physiological processes, including platelet aggregation, memory formation, sexual function, blood pressure homeostasis, and immune response. Dysfunction along the NO/GC- 1/cGMP pathway is a crucial component in multiple cardiovascular diseases, namely pulmonary hypertension, insufficient platelet aggregation, heart failure, and stroke. Therefore, GC-1 is heavily targeted for drug discovery. Stimulation of the NO signaling pathway has been used therapeutically to treat various forms of cardiovascular disease for more than a century, beginning with the administration of amyl nitrite and nitroglycerin to relieve symptoms of angina pectoris. Despite its significance to cardiovascular health and disease, the structure, regulation, and function of GC-1 remains poorly understood. This work describes the solvent accessibility of GC- 1 using hydroxyl radical footprinting coupled with tandem mass spectrometry, as well as the effect of phosphorylation sites along full-length GC-1
