| مستخلص: | In this thesis, we develop experimental approaches to investigate the folding of DNA origami objects and to extract the kinetics of the self-assembly process. We present two fluorometric assays which allow observing the binding of individual DNA strands of the DNA origami in real-time, revealing the sequence of binding events within the self-assembly process. A detailed analysis connects the strand properties and the strand routing in the design with the observed folding pathway. In dieser Arbeit entwickeln wir experimentelle Ansätze, um die DNA-Origami-Faltung zu untersuchen, sowie die Kinetik der Selbstassemblierung zu extrahieren. Zwei fluorometrische Ansätze ermöglichen die Beobachtung der Bindung einzelner Stränge des DNA-Origamis in Echtzeit und machen die Reihenfolge der Bindungsereignisse innerhalb der Selbstassemblierung zugänglich. Eine detaillierte Analyse verbindet die Eigenschaften der Stränge und ihr Routing mit dem beobachteten Faltungspfad. |
