Influenza virus, which causes annual epidemics of respiratory infection, is associated with significant morbidity and mortality. Lately, reports of emerging resistant viruses against Zanamivir and Oseltamivir have begun to emerge of viruses resistant to antiviral drugs (i.e. Zanamivir and Oseltamivir), which have been useful in the treatment of influenza virus infection. In the meantime, the developing of new effective drugs has been necessary. In this study, we investigate the antiviral mechanism of a Chinese herbal medicine CGU-V002. Antiviral neutralization (inhibition of virus-induced cell death) screening demonstrated that CGU-V002 is an effective anti-influenza virus agent. CGU-V002 exhibits an EC50 of 0.016 ± 0.005 mg/mL against influenza virus A/WSN/33 (H1N1), with a broad-spectrum of inhibitory activity against different strains of human influenza A viruses, including clinical Oseltamivir-resistant viruses and influenza B strains, but not with enteroviruses. According to the result of time-of-addition assay, that CGU-V002 inhibits virus replication at the early, middle and late stages. Further perform different assays to address the inhibitory activities of CGU-V002. On the viral early stage, that CGU-V002 inhibits viral hemagglutination, attachment and penetration to cells. On the middle stage, that CGU-V002 decrease viral RNA and protein synthesis, but no significant difference in its protein distribution. In addition, on the late stage, CGU-V002 partially inhibits viral NA hydrolysis activity. In summary, CGU-V002 effectively inhibits virus replication, which may exert its anti-viral effect through inhibiting virus entry.
