Study on Using HPLC to Determine the Heat-induced Products from Lactose Chin-Ming Hsu Abstract Lactose can produce lactulose and galactose when lactose are in thermal environment. Products of heat — induced browning products responding with protein and loss nutrition, when lactose and protein are together in thermal environment. The study uses galactose, glucose, lactulose, lactose and lysine as standard samples, each of whose concentration is 4.8%. The HPLC method using a NC — 04 column, and elute by solventⅠ: the concentration of acetonitrile is 75% and flow rate is 1.2mL/min or solventⅡ: mixing acetonitrile, acetone with water, their ratio of volume is 3:3:2 and flow rate is 1.0 mL/min. The best efficiency of separation can be got at the two types of solvent and the concentration of the sacchairides above have to be set up. After that, mix lactose with lysine as samples and heat them with 63℃,30、60、90 min;75℃,15、30、45 sec;135℃,4、8、12 sec or 121℃,20、40、60 min. After heating treatment, inject the samples to the HPLC and observe the change of carbohydrates and lysine in the samples. As the result, the more hard thermal processes, the lower amount of lactose and lysine, while the higher amount of galactose and lactulose, when lactose or lysine is heated separately. The HPLC analyses show that lactulose can be detected in the samples are treated at 63℃,90 min;135℃,12 sec and 121℃,20、40、60 min, when mixing lactose with lysine and heating them at the same time.
