الوصف: | Introduction and Aims: Angiotensin (Ang)-(1-7) acts via the G protein-coupled receptor Mas to oppose Ang II/AT1 receptor-mediated effects on cell proliferation and cardiovascular homeostasis. In the present study, we examined the role of the Mas receptor in early UB branching morphogenesis and GDNF/Ret/Wnt11 pathway gene expression. Methods: Paired CD1 mice metanephroi were dissected on embryonic (E) day E12.5 and grown on polycarbonate transwell filters located on top of DMEM/F12 medium alone (control, n=5) or in the presence of the specific Mas receptor antagonist A-779 (10-6 M, n=5) for 24 hours. In addition, we examined UB branching in metanephroi from Mas +/+ (n=4) and Mas -/- (n=4) mice on E13.5. The UB epithelia were visualized by staining the metanephroi with anti- pancytokeratin antibody (1:200, Sigma) and the absolute number of UB tips in each group was counted. Results: A-779 increased the number of UB tips compared to control at 24 (41.2±2.7 vs. 31.2±1.9; p<0.05). Similar increase in the UB tip number was observed in Mas-/- compared with Mas+/+ kidneys (43.1±1.5 vs. 34.7±1.3; p<0.01). To determine whether the observed induction of UB branching by A-779 is due to upregulation of the GDNF/Ret/Wnt11 morphogenetic pathway, we measured GDNF, Ret and Wnt11 mRNA levels after 24 hours of culture by real-time quantitative PCR. Treatment with A-779 (n=3) significantly increased GDNF (1.88±0.2 vs. 1.0±0.0; p<0.05), Ret (1.62±0.1 vs. 1.0±0.0; p<0.01) and Wnt11 (1.91±0.1 vs. 1.0±0.0; p<0.01) mRNA levels in whole metanephroi compared to control (n=3). In contrast, A-779 decreased expression of Spry1 (0.32±0.02 vs. 1.0±0.0; p<0.01), an endogenous inhibitor of the GDNF/Ret/Wnt11 pathway. Conclusions: These findings indicate that endogenous Ang-(1-7) and its Mas receptor counteract the effects of Ang II/AT1 receptor axis on UB branching and GDNF/Ret/Wnt11 pathway gene expression.
