الوصف: | The porcine reproductive and respiratory syndrome virus (PRRSV) and the swine influenza A virus (swIAV) are leading players of the porcine respiratory disease complexe. While much is known about PRRSV and swIAV single-infections, the possible interactions between these two pathogens are still misunderstood. Moreover, the modified live vaccines (MLV) against PRRSV are injected to piglets when swIAV infection may occur. The objectives of this thesis project were to study the clinical, virological and immunological outcomes of PRRSV/swIAV co-infection in pigs and to investigate the effect of swIAV infection on MLV vaccination. We showed that a swIAV infection reduces PRRSV multiplication in lungs, delays MLV viremia without interfering with vaccine protection and accelerates the induction of the cell-mediated response specific to PRRSV. In addition, PRRSV pre-infection attenuates the clinical signs, inflammatory and antiviral responses linked to influenza infection, and increases the anti-swIAV antibodies levels (IgG, IgA, anti-HA and neutralizing) in lungs. Correlation analyses showed that the induction of the interferon type I pathway, which is blocked by PRRSV but stimulated by swIAV, appears to play a key role in this bidirectional viral interference. The study of underlying mechanisms has been initiated through co-infection experiments in a co-culture system of alveolar macrophages and tracheal epithelial cells. The new knowledge acquired confirms the importance of determining the etiology of respiratory syndromes, in order to improve the control measures (including vaccination protocols) by taking into account all the pathogens involved. ; Les virus du syndrome dysgénésique et respiratoire porcin (SDRPV) et de l’influenza A porcin (swIAV) sont deux acteurs majeurs du complexe respiratoire porcin.
