Growth at the shoot apical meristem (SAM) is essential for shoot architectureconstruction. The phytohormones gibberellins (GA) play a pivotal role incoordinating plant growth, but their role in the SAM remainsmostly unknown.Here, we developed a ratiometric GA signaling biosensor by engineering oneof the DELLA proteins, to suppress its master regulatory function in GA transcriptionalresponses while preserving its degradation upon GA sensing. Wedemonstrate that this degradation-based biosensor accurately reports oncellular changes inGA levels and perception during development.Weused thisbiosensor to map GA signaling activity in the SAM. We show that high GAsignaling is found primarily in cells located between organ primordia that arethe precursors of internodes. By gain- and loss-of-function approaches, wefurther demonstrate that GAs regulate cell division plane orientation toestablish the typical cellular organization of internodes, thus contributing tointernode speci fi cation in the SAM. |
