الوصف: International audience ; Introduction and aim of the study: Gestational diabetes (GDM) is a major public health issue, affecting 1 in 6 pregnancies worldwide. GDM increases the offspring's risk of developing type 2 diabetes (T2D) in adulthood. Epidemiological studies show the benefits of breastfeeding duration on the child's glycemic status, but the long-term effects are still debated. Preliminary results suggest an adaptation of milk composition to maternal hyperglycemia. We therefore hypothesize that maternal hyperglycemia may modify the composition of milk in terms of key regulators of insulin sensitivity, with repercussions on the metabolism of the offspring. To test this hypothesis, cross-adoptions were performed in a nutritional model of DG rats. Material and methods: Female rats were fed a high-fat, high-sucrose diet (HFHS) or a standard diet (CTL) one week before and during 3 weeks' gestation. During the 3-week lactation period, the HFHS group was fed a standard diet (4HFHS) or maintained on HFHS (7HFHS). We calculated the area under the curve (AUC) and an insulin sensitivity index (ISIGutt) following oral glucose tolerance tests (OGTT) performed during gestation (G12) and lactation (L12). Milk composition was characterized on day 1 of lactation (L1), then at L8, L14 and L18 by targeted analysis of total fatty acid profiles (gas chromatography), or amino acid and sphingolipid concentrations and untargeted analyses of lipidomic profiles (liquid chromatography/tandem mass spectrometry). Plasma lipidome/metabolome phenotyping of newborn pups (D0) was performed using Biocrates' MxP500 quant kit on the Sciex QTRAP 6500+. Then, following cross-adoptions with normoglycemic or hyperglycemic dams, postnatal monitoring of offspring was carried out. Insulin sensitivity of adult offspring was measured (hyperinsulinemic euglycemic clamps), as well as insulin secretion capacity (OGTT) in response to a hypercaloric challenge from D120 to D220.
