الوصف: Pigment epithelium‐derived factor (PEDF) is a multifunctional protein with important roles in regulation of inflammation and angiogenesis. It is produced by various cell types including endothelial cells (EC). However, the cell autonomous role PEDF plays in regulation of EC function remains elusive. Here we prepared lung EC from wild type (PEDF +/+) and PEDF‐deficient (PEDF−/−) mice and determined the impact of PEDF‐deficiency on EC function. We show that PEDF−/− EC are more migratory and fail to undergo capillary morphogenesis in Matrigel compared to PEDF+/+ EC. Although no significant differences were observed in the rates of apoptosis of PEDF−/− EC compared to PEDF+/+ EC, PEDF−/− EC proliferated at a slower rate. PEDF−/− EC also expressed increased levels of proinflammatory markers including VEGF, iNOS, VCAM‐1, as well as exhibiting altered cellular junctional organization. Lack of PEDF expression resulted in increased adhesion of EC to extracellular matrix proteins, including collagen IV, fibronectin and vitronectin. These changes were associated with decreased expression of α1, α6 and α7 integrins in PEDF−/− EC. PEDF−/− EC also expressed decreased levels of thrombospondin‐2, tenascin‐C and osteopontin while that of fibronectin was increased. Together our data suggest that PEDF modulates EC pro‐inflammatory and pro‐angiogenic properties by suppressing the inflammatory phenotype of EC. (NIH/EY18179, EY21357, ADA/1‐10‐BS‐160)
