الوصف: | Quorum sensing serves as a communication technique utilized by bacteria to monitor the cell density in their environment. These quorum sensing systems have attracted a lot of attention as building blocks in the construction of genetic circuits, allowing the design of population-level regulated circuits, such as noise regulation or the creation of consortia comprising communicating bacterial strains. Moreover, these systems offer great possibilities to optimize pathway regulation by allowing the auto-inducible expression of a gene or pathway of interest, thereby circumventing the necessity of expensive inducers, such as IPTG, to induce a pathway at higher cell densities. In this research, two different quorum sensing systems, the LasR/LasI and EsaR/EsaI system, were researched. The systems were investigated and engineered for limited crosstalk facilitating their concurrent application within the same bacterial strain. Additionally, a novel promoter was created that is only activated in the presence of the auto-inducer molecules of both quorum sensing systems. This offers numerous new possibilities for genetic circuits outputs, with promising applications for pathway regulation. For example, a multistage switch, consisting of a growth stage followed by two production stages separated in time, holds great promise for the regulation of complex pathways competing with the central metabolism or exerting toxic effects. |
