| الوصف: | (A) The pGL3-basic (negative control), pGL3-1146/-646 (positive control), and pRL-TK (internal control) were co-transfected into MG-63 cells. Transfected cells were incubated in the presence (solid bar) and absence (open bar) of 10% FBS with LY294002 (10 μM), GŐ6983 (100 nM), SB203580 (20 μM), SP600125 (10 μM), U0126 (10 μM), and Compound C (10 μM) inhibitors for 24 h. Relative luciferase activity was normalized with the Renilla luciferase activity derived from pRL-TK. MG-63 cells were incubated in the presence and absence of 10% FBS with SB203580 (20 μM) for 24 h and analyzed by RT-PCR (B) and quantitative real-time PCR (C). Data represent mean ± SEM for three independent experiments with triplicate measurements. * P < 0.05; ** P < 0.01, *** P < 0.001. |
