The neurodegeneration seen in Alzheimer's disease (AD) is largely attributed to the over‐production and accumulation of the protein Amyloid beta (Aβ), which becomes toxic to the cells and induces dysfunction of cellular organelles to inflict the damage. Recent publications have demonstrated that the epoxide derivatives of arachidonic acid, epoxyeicosatrienoic acids (EETs; 14,15‐, 11,12‐, 8,9‐ and 5,6‐EET), are cytoprotective in the brain and promote neuronal growth. The involvement of EETs in AD has not been studied yet. Our previous data suggests that Aβ affects EETs production (Sarkar et al, Neuroscience, 2011, p241–249). In the current study, we evaluated the cytoprotective potential of exogenous EETs against Aβ damage in cultured astrocytes. Pre‐incubation with 14,15‐ and 11,12‐EET protected the cells against Aβ‐induced mitochondrial fragmentation and membrane depolarization. Moreover, EETs reversed the Aβ‐mediated uncoupling of electron transport chain from ATP synthesis and reduced cellular ROS production. Our data provides the first evidence of the involvement of EETs in the downstream pathway of Aβ toxicity and manipulating this signaling cascade may improve AD pathology.
