Zymoseptoria tritici is the major fungal pathogen of wheat in Europe. Despite its significance, very little is known about how Z. tritici is able to induce disease and how it evades host defenses during its extended latent/biotrophic growth phase inside the wheat leaf. Aside from well-characterized LysM domain effectors that mask the fungus’ chitin from the host, Z. tritici’s mechanisms of host immune suppression are poorly understood. We have performed different screens of Z. tritici effectors in both Nicotiana benthamiana and wheat, and have identified effector candidates as immune-suppressors. To date, we have primarily focused on effectors able to suppress BAK1-triggered immunity. Z. tritici has two homologues of the BAK1-interacting effector NIS1. One of these homologues is highly expressed during the biotrophic growth phase, and so we have used it as a positive control for immune suppression. At this stage, we have identified an additional four putative BAK1-suppressing effectors among the first 12 effectors screened. Each of these effectors are capable of suppressing Pseudomonas syringae-induced cell-death in N. benthamiana. In summary, our studies have identified novel effectors used by Z. tritici to suppress host immune responses. This is part of an ongoing project to dissect the wheat-Z. tritici molecular interactions, with a long-term goal of identifying mechanisms of host-specificity. |
