الوصف: | To better understand bacterial infections in the context of pulmonary infections in cystic fibrosis patients, it is important to better understand the interactions between pathogens. We looked at the interactions between Pseudomonas aeruginosa, a pathogen that chronically infects cystic fibrosis patients, and other pathogens found in cystic fibrosis patients through the battle between these bacteria for iron supply. Iron is a key element for bacterial growth, but it is very poorly bioavailable in host fluids. The bacteria then produce siderophores to bind iron in the extracellular medium and allow its import into the bacteria. P. aeruginosa produces two siderophores, pyoverdine and pyochelin. During chronic P. aeruginosa infection, the production of these siderophores varies, along with the virulence of the strains. To better understand the importance of siderophores for iron import and for these virulence mechanisms, we studied the interactions between P. aeruginosa isolates from a single patient, but collected at different times during chronic infection, and 5 other pathogens. We first better characterised the regulation of pyochelin production, which is less well studied, and then developed a tool to differentiate the bacteria in these co-cultures using a fluorescent marker. We were thus able to better specify the importance of these siderophores as virulence factors and as iron import factors. The decrease in their production during chronic infection allows other bacteria to proliferate better and bacterial interactions to be established. Finally, we also established a link between certain antibiotic treatments, recurrent in these patients, and the evolution of siderophore production in P. aeruginosa. ; Afin de mieux lutter contre les infections bactériennes dans le cadre des infections pulmonaires chez les personnes atteintes de mucoviscidose, il est important de mieux comprendre les interactions entre pathogènes.
