الوصف: | International audience ; Epidemiological evidence suggests that foetal hyperglycaemia in gestational diabetes (GDM) context programs offspring susceptibility to type-2-diabetes. Emerging benefit effects of breastfeeding duration on child's metabolic status were reported beside changes in GDM-breast-milk (BM) composition. We investigated whether maternal hyperglycaemia impacts BM content in insulin-sensitivity regulators and their effects on offspring homeostasis using a pre-clinical cross-fostering rodent model combined with an integrative metabolomic approach. Female rats were fed high-fat high-sucrose diet (HFHS) or standard diet (CTL) one week before and during gestation. During lactation, HFHS group was fed standard diet (4HFHS) or maintained on HFHS (7HFHS). Oral glucose tolerance tests (OGTT) were performed during gestation (G12) and lactation (L12). Milk metabolome/lipidome was characterized throughout lactation period (L1/L8/L14/L19). Offspring insulin sensitivity was studied by OGTT (D60/D210) and euglycemic-hyperinsulinemic clamps (D200) in response to hypercaloric challenge (D120 to D220).At G12, HFHS dams showed lower insulin-sensitivity vs CTL, which was maintained in 7HFHS vs 4HFHS and CTL dams at L12. Throughout lactation, 7HFHS vs CTL milk showed an increase in insulin-trophic amino acids and ω6/ω3 polyunsaturated fatty acids ratio, known as insulin/incretin secretagogues, but also in various sphingolipids. At D220, offspring born to HFHS dams and suckled by CTL dams showed hyperglycemia (in males) and an insulin-sensitivity lower trend (in females) in response to hypercaloric challenge. Gestation and lactation hyperglycemia induce BM-adaptations associated to a gender-metabolic flexibility on offspring. We are currently analysing longitudinal offspring blood metabolome/lipidome phenotyping data in a specific manner depending to be nursed by euglycemic vs normoglycemic dams. |
