الوصف: Maslačak je biljka koju odlikuje visoka koncentracija fenolnih spojeva te može poslužiti kao dobra sirovina za njihovu ekstrakciju. Ekstrakcija potpomognuta visokim hidrostatskim tlakom (HPAE) i ubrzana ekstrakcija otapalima pri povišenom tlaku (ASE) su novije metode izolacije bioaktivnih komponenti iz biljnog materijala. Cilj ovog rada je bio ispitati utjecaj odabranih parametara na izolaciju fenolnih spojeva iz lista i korijena maslačka. Optimalni uvjeti za izolaciju iz lista HPAE metodom su 70 % - tni etanol kao ekstrakcijsko otapalo, temperatura 60 °C, tlak 500 MPa i vrijeme ekstrakcije 5 minuta, a za izolaciju iz korijena50 % - tna vodena otopina etanola kao ekstrakcijsko otapalo, temperatura 60 °C, tlak 500 MPa i vrijeme ekstrakcije 5 minuta. Optimalni uvjeti za izolaciju iz lista ASE metodom su 50 % - tna vodena otopina etanola, temperatura 60 °C i vrijeme ekstrakcije 10 minuta, a iz korijena 50 % - tna vodena otopina etanola, temperatura 80 °C i vrijeme ekstrakcije 15 minuta. Primjenom ASE metode dobiveni su veći prinosi fenolnih spojeva nego HPAE metodom. Dandelion is characterized by high concentrations of phenolic compounds and it can serve as a good raw material for their extraction. High hydrostatic pressure assisted extraction (HPAE) and accelerated solvent extraction (ASE) are relatively new technics which are used for the extraction of bioactive compounds from plant materials. The aim of this study was to investigate the influence of different parameters on the isolation of phenolic compounds from the leaves and roots of dandelion. Optimum conditions for extraction of an entire amount of phenols from leaves using HPAE method was as follows: ethanol in polarity of 70 % with 5 minutes duration at 60°C and 500 MPa pressure, on the other hand for extraction from roots, optimum conditions were: ethanol in polarity of 50 %, with 5 minutes duration at 60°C and 500 MPa pressure. Optimum conditions for extraction of an entire amount of phenols from leaves using ASE method was as follows: ethanol in polarity of 50 % with 10 minutes duration at 60°C. On the other hand, for extraction from roots optimum conditions were: ethanol in polarity of 50 %, with 15 minutes duration at 80°C. By applying ASE method higher yields of phenolic compounds were obtained than with HPAE method.
