The techniques employed on the DNA identification bear two advantages upon the conventional ones: the chemical stabilization of the DNA, even after a long period of time and its occurrence in all the nucleated cells of the human organism, which allows to convict or acquit a suspect with a drop of blood or an unique hair found in the crime scene. Forensic evidence, from tissues embedded in paraffin archived and safeguarded on The Pathology and Genetics Institute are, in some cases, the only register available of the collected sample. However, there is still a few good protocols in forensic context, to recover DNA from this kind of material, usually from old samples and human remains. The aim of this study is to evaluate methods of extraction and purification of the DNA, using biological archival materials, in order to determine a protocol that offers good recovery of the DNA and adapts on the Institute of Research and Expertise in Forensic Genetics (IPPGF), forensic laboratorial reality and others forensic services. Twenty two samples from cadaver tissues, included in paraffin blocks were submitted to two pre treatment methods: deparaffination using xylene and heating in microwave. After that, four other methods were used for DNA extraction, that includes: Organic Method, Chelex 100®, Purification on membranes (NucleoSpin) and the Resin purification method (DNA IQ). We observed that the spectrofometrical results are not good parameters to support and direct the technical procedures of extraction from the material embedded in paraffin archives, because even when the spectrofometrical results revealed the presence of the DNA, there was no successful amplification of the expected fragment. The polymerase chain reaction demonstrated to be an efficient technique, to amplify a degraded DNA in low quantity. The best DNA concentration for PCR TR were obtained in organic (phenol-chloroform-isoamyl alcohol) and Nucleospin metodologies, been subsequently paired for comparison, using the non parametrical Friedman test, that showed significant differences (p
