Abstract (English): The bacteria with high-yield esterase were screened from Daqu, and identified by morphological identification and molecular biological i- dentification. The excellent mutant strains were selected by atmospheric and room temperature plasma-ultraviolet (ARTP-UV) compound mutagenesis, and the genetic stability test was carried out to improve the esterase production ability. The results showed that a bacterial strain S3-51 with esterase activity of (13.73±0.04) U/ml was screened from strong-flavor (Nongxiangxing) Daqu, which was identified as Sphingomonas paucimobilis. The mutant strain ARUV3-2 with the highest enzyme activity was screened by ARTP-UV compound mutagenesis of strain S3-51. After genetic stability test, the esterase activity of mutant strain ARUV3-2 was stable at (17.42±0.32) U/ml, which was 26.88% higher than that of the preliminarily screened strain. The mutant strain ARUV3-2 provided potential for the industrial synthesis of fatty acid ethyl esters and the improvement of Baijiu quality. [ABSTRACT FROM AUTHOR]
Abstract (Chinese): 该研究从大曲中筛选出高产酯酶的细菌, 对其进行形态学鉴定和分子生物学鉴定, 利用常压室温等离子体-紫外(ARTP-UV) 复合诱变选育出优良突变株, 并进行遗传稳定性试验, 提高其产酶能力。结果表明, 从某浓香型酒厂大曲中筛选出一株产酯酶酶活力 为(13.73±0.04) U/mL的细菌, 编号为S3-51, 其被鉴定为少见鞘氨醇单胞菌(Sphingomonas paucimobilis)。对菌株S3-51进行ARTP-UV 复合诱变, 选育出酶活最高的突变株ARUV3-2, 经遗传稳定性测试后, 菌株ARUV3-2的酯酶酶活力稳定在(17.42±0.32) U/mL, 相比于 初筛时的产量提高了26.88%。菌株ARUV3-2为脂肪酸乙酯类化合物的工业合成和白酒质量的提升提供潜力。 [ABSTRACT FROM AUTHOR]
