مستخلص: Ardisia humilis Vahl is one of the natural plants, popularly known as Lampeni in Indonesia. This plant has been highlighted to perform health-beneficial activities such as antioxidants. This study aimed to investigate the effect of extraction with 70% ethanol and its fractions on total phenolic content (TPC), total flavonoid content (TFC), and 2,2′-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. Seventy percent of ethanol followed with another solvent such as hexane, ethyl acetate, or water was used to extract Ardisia humilis Vahl Leaves. The extracts, next, were evaluated for TPC, TFC, and DPPH radical scavenging assay. The result of the study showed that ethyl acetate fraction compared to other extracts showed the highest TPC and TFC (P< 0.05), showing 209.17±11.9 mgGAE/g extract and 83.08±6.10 mgQE/g extract, respectively. Moreover, 70% ethanol, ethyl acetate fraction, and water fraction extracts performed an equal radical scavenging activity (P >0.05), showing IC50 of 29.26±0.28 ppm, 24.69±1.36 ppm, and 25.40±3.98 ppm, respectively, but ascorbic acid (IC50=13.35±0.37 ppm) was significantly the highest rather than all of the extracts (P< 0.01). In addition, the hexane fraction was shown as having the lowest TPC, and IC50 value, resulting in 32.73±5.17mgGAE/g extract, and 120.02±7.62 ppm. In conclusion, the experiment revealed that the ethyl acetate fraction of Ardisia humilis Vahl extract comprises a high amount of flavonoid and phenolic compound, contributing to its potency as a very strong antioxidant agent.
