مصطلحات موضوعية: INTRODUCTION & OBJECTIVES: The vascular endothelial growth factor (VEGF) is a major mediator of angiogenesis involving tumor growth and metastasis. Polymorphisms in VEGF gene may regulate VEGF production. In this case-control study, we investigated whether functional polymorphisms (-460C/T, +405C/G,+936C/T) in the VEGF gene are associated with the higher risk of developing a bladder cancer. MATERIAL & METHODS: VEGF genotypes and haplotypes were determined in 46 patients 36 male and 10 female, medium age 70 (range 51 – 91) with bladder cancer and 100 healthy controls. 42 patients have no muscle invasive bladder cancer, (29 high grade and 13 low grade) only 4 patients present muscle invasive bladder tumour. RESULTS: There were no associations between –460C/T and +405 C/G genotypes and the risk of bladder cancer. The combined +936 TT+ CT genotype was significantly associated with increased risk of cancer (OR 2.16, % CI 0.97-4.85, p=0.034). In haplotypes analysis the –460T/+405G/+936T haplotype was significantly (p=0.001) associated with increased risk of bladder cancer when compared with the most common CGC haplotype (OR 5.38, %CI 2.28-12.81). CONCLUSIONS: In conclusion, VEGF polymorphism in our study was involved in bladder cancer susceptibility. Further work is required to replicate these findings in a larger population and to correlate VEGF polymorphism with progression and recurrence of bladder cancer, Settore MED/24 - Urologia
